General description
Potato Dextrose Agar is recommended for the isolation and enumeration of yeasts and molds in dairy and food products. Potato extract serves as a source of carbon, nitrogen, minerals, vitamins, and other essential growth nutrients. Dextrose acts as a source of carbohydrates. Agar is the solidifying agent. The accompanying bacterial flora is suppressed by the low pH of 3.5. The growth of yeasts and molds is promoted in this medium, and the fungi develop typical morphology. The addition of Rose Bengal further suppresses the growth of bacteria and restricts the size and spreading of mold colonies.
Application
Potato Dextrose Agar is used for the isolation and enumeration of yeasts and molds in dairy and food products. The media is also used for stimulating sporulation, maintaining stock cultures of certain dermatophytes, and as a differential media for certain varieties of dermatophytes on the basis of pigment production.

Preparation Note
Suspend 39 g in 1 litre of distilled water. Bring to the boil to dissolve completely. Sterilize by autoclaving at 121°C for 15 minutes. In order to suppress bacterial growth, 1 ml of 10% sterile lactic acid can be added for dropping pH to 3.5. Mix well before pouring.
